Khouma Babacar opened the scoring before Marco Marchionni made it 2-0 prior to half-time interval. Alessio Cerci extended the home side's advantage only a few minutes after the restart.
The Viola meant business from the start with Babacar the first to react to Marchionni's thunderbolt from the edge of the area after only 30 seconds, almost looping his shot over the advancing Pietro Marino.
The visitors had a chance in the 16th minute when Simone Rizzato combined with Dominic Adiyiah on the left, who then squared a pass for Domenico Danti, but his shot fizzled across goal. The Amaranto were on the attack again a minute later, as Vlada Avramov's poor goal kick fell to Danti, who whipped in a ball for Adiyiah in the box, but he could not turn and shoot on target, flashing his effort just wide.
Babacar then put the hosts up after 28 minutes, as a simple pass from Federico Carraro reached the Senegalese, and unmarked, he slotted home to make it 1-0.
Cristiano Zanetti wanted to get on the scoresheet himself, and his chance came just after the half-hour, as he was allowed some space in front of goal, but his attempt went wide of the right post.
Marchionni extended the Viola's lead with two minutes left in the first-half, as he was left unmarked at the back-post from a Carraro corner-kick, and from only a yard out, blasted the ball into the roof of the net to make it 2-0.
Two minutes after the restart the score was 3-0, as Alessio Cerci rolled his shot past the advancing goalkeeper.
Haris Seferovic came close to further extending the lead when the substitute turned and fired his attempt just wide.
Reggina's best effort came from an Ivan Castiglia free-kick from 30 yards out on 73 minutes. Aiming for the top right-hand corner and curling wickedly, Avramov tipped it away, outstretched to his left.
Giacomo Tedesco was shown a straight red card for his late sliding challenge on Gianluca Comotto with seven minutes to go.
A fourth goal was almost added as Zanetti's flick found Seferovic, who chipped the advancing 'keeper, but Dani Verruschi booted the bouncing ball off the line.
La Viola will now take on Parma in the last 16.
