A late goal by Kwasi Donsu sealed the winner for the hosts who faced pressure from the visitors all through second half due to a numerical disadvantage.
Aduana coach Yussif Abubakar made three changes to the squad that started against Raja Casablanca in Caf Confederation Cup last week.
Yahaya Mohammed, Charles Boateng and goalkeeper Joseph Gordon came in while the hosts paraded a squad which had Kwesi Donsu and Kwame Boateng leading the attack.
After a slow start, a nice build up play involving Justice Blay saw Kwasi Donsu curling his effort into the right bottom corner to hand Medeama the lead by the 5th minute.
By the 17th minute, Medeama's Bright Enchil made an attempt to push his way into Aduana's box but Hafiz Adams prevented him.
Four minutes later, the hosts came close once more after Boateng connected a teeing cross from Eric Kwakwa, who released Agyenim Boateng but he failed to find the target.
On 33rd minute, Aduana defender Hafiz wasted a close range effort, while Aidoo got injured in the build up and had to be replaced by Emmanuel Akuoko.
Aduana came close to equalising by the 47th minute but Emmanuel Boateng's effort was saved by goalkeeper Eric Ofori Antwi.
However, the Dormaa Ahenkro-based club pulled level through Sam Adams, who transformed a spot kick following a foul on Yahaya Mohammed.
Under pressure from the visitors, Medeama goalkeeper Antwi handled the ball outside his area and was shown a straight red card.
However, Medeama were able to walk away the happier side despite finishing the encounter with a man less. Donsu bent the last free-kick of the game straight into the net to move Medeama to top of the log for the first time this season with 23 points, two above Ashanti Gold.
