The game was expected to be played on November 10, but was postponed due to heavy rainfall and rescheduled for Sunday, pending an improvement in weather conditions.
La Bombonera is the stage for the opening joust between the Superclasico foes as they compete for the right to be crowned champions of South America.
Meetings between the sides are typically tempestuous and they will both be desperate to lay down a marker ahead of the second leg, which takes place at El Monumental two weeks later on November 24.
Boca have a better history in the competition than River, having won the trophy on six previous occasions, but the visitors can take a step towards closing that gap on Saturday by inching closer to their fourth crown.

Betting & Match Odds
Boca are favourites to win the first leg at La Bombonera, with bet365 giving odds of 7/5. River's chances of repeating their September feat are rated 12/5 and a draw is a 19/10 bet.
The whole world will be tuning in on Sunday for a glimpse of one of the most intense football rivalries in football as Boca and River lock horns for the right to get their hands on the Copa Libertadores.
Such is the vitriol associated with the Superclasico in Argentina that neither La Bombonera nor El Monumental will feature visiting supporters, meaning the River players are truly entering the gauntlet this weekend - just as Boca will do in the second leg on November 24.
While they are undoubtedly Argentina's most successful clubs of all time with 69 league titles between them, both sides have made disappointing starts to the 2018-19 campaign, finding themselves mid-table after the opening 11 Superliga matches, with River - whose form has been particularly bad - in 10th place, two points behind eighth-placed Boca.
However, despite trailing Boca in the league, River may actually boast the psychological edge over their rivals after striking first blood back in September, escaping La Bombonera with a 2-0 win, thanks to goals from Gonzalo Martinez and Ignacio Scocco.
Nevertheless, Boca will have a partisan crowd of 49,000 roaring them on in search of revenge and - more importantly - glory.
