Coach Takeshi Okada is keen to take on quality opposition before next June especially as a March 3 friendly against Portugal has been cancelled.
Japan will take on Denmark, the Netherlands and Cameroon in South Africa but are happy to take on South American opposition.
"Teams from South America always approach matches wanting to win. This game will help toughen Japan up,” Okada told reporters in Tokyo.
Samurai Blue are also hoping to arrange games against top class sides in April and May.
Japan also take on China, South Korea and Hong Kong in February in the 2010 East Asian Championship.
