A partially torn ankle ligament forced the midfielder from the field of play during BVB's 1-0 defeat to Bayer Leverkusen, while Nuri Sahin also had his afternoon ended prematurely with an ankle problem.
The pair join Mats Hummels, Neven Subotic, Ilkay Gundogan and Marcel Schmelzer on the sidelines as injuries threaten to seriously undermine BVB's season.
Sahin remains a 50/50 chance to start the game ahead of the must-win clash, with Dortmund needing a win to have any hope of qualifying for the knockout stages of the competition.
Speaking after the defeat, coach Jurgen Klopp told reporters: "We lost 1-0, but that's the least of our problems today."
The defeat to Leverkusen leaves Dortmund six points adrift of Sami Hyypia's men in third place and 10 behind leaders Bayern Munich in the Bundesliga.
