The Blaugrana stars have had their commitment to the Spain cause questioned after they were allowed to go back to the Catalan capital instead of accompanying the squad for the midweek fixture.
Puyol picked up a booking in the win over Armenia and is suspended, while Xavi has a slight injury that will be examined by Barca club medics.
"If anyone doubts their commitment then they have to then doubt everyone's commitment," Busquets was quoted as saying by El Mundo Deportivo.
"Puyol is suspended because of [yellow] cards and it is normal [that he returns home] because he cannot play.
"Xavi, like [Valencia's Carlos] Marchena, has a knock and should wait for the test results before evaluating things.
"It is a decision that the coach takes with the coaching staff and the player. Nobody should ever doubt the players' commitment.
"They would only ever miss games through suspension or injury, or like Cesc's case as his grandfather is ill."
