The 26-year-old has been at Borussia Dortmund since 2008 and has made 74 appearances for the club.
Many German clubs have been linked with a move for the defender, with Bayer Leverkusen, Borussia Monchengladbach and Hannover all reportedly interested in securing Santana's signature.
Wolfsburg are believed to be waiting to see whether Roma complete the signing of Simon Kjaer, who was on loan at the Italian side from the 2009 Bundesliga champions, before making a final decision.
Should Kjaer's move go ahead, Santana would be signed as a direct replacement.
Wolfsburg finished in 8th position in the Bundesliga this season.
