The Italy international has started all four of their games so far this season, and he is hoping to witness more action against the champions.
Captain Alessandro Del Piero recently said the game is not decisive, but Marchisio has a different opinion.
"It is the most important game for us on paper to date, we are playing against the Italian champions and we know it is a great chance for us to take advantage," he is quoted as saying by La Repubblica.
"We want to win the game so we can stay on top of the league.
"Milan are a great team and so it is very important for us to get it right."
Kick-off at the Juventus Arena is at 20:45 (CET). Juventus' Fabio Quagliarella is in contention after for a starting place after he featured in a youth game in midweek following his recent return from long-term injury as a substitute in the 1-1 draw with Catania last weekend.
