Michel Preud'homme's men are now three points clear of their nearest rivals PSV, who can leapfrog Twente in pole position on goal difference if they defeat Feyenoord on Sunday.
Den Haag and Twente went into the half-time break with neither side managing to find the target, although the visitors thought that they had opened the scoring just before the break when Bryan Ruiz's effort ended up in the back of net. However, the goal was chalked off for offside.
Ruiz continued to cause problems for the home team and eventually broke the deadlock just after the hour-mark, receiving the ball from Nacer Chadli before slotting it past the helpless Gino Coutinho.
However, the lead was canceled out in the 70th minute as Dmitri Bulykin's volley went in - it was the Russian striker's 19th goal of the season.
Twente regained their lead in the 86th minute when De Jong scored what proved to be the winning goal, pouncing on the rebound after Coutinho saved his initial header.
Next up for the champions is a home encounter against Willem ll while ADO Den Haag will continue their pursuit for European football when they entertain Groningen next week.
