The Boys in Green manager has included Norwich City midfielder Wes Hoolahan in the squad after running the rule over the player at Carrow Road.
Captain Robbie Keane has not been included in the squad, as he will be involved in a crucial playoff series with LA Galaxy, while West Brom striker Shane Long is also not named in the 26-man panel, after suffering a strained hamstring.
First choice defensive pair Richard Dunne and Sean St Ledger miss out due to injury. However, Everton defender Shane Duffy and Chelsea midfielder Conor Clifford are set to win their first senior caps after being included.
Speaking at the announcement, Trapattoni said: "I have decided to choose a provisional 26 man squad for this game due to injury doubts over players. As we have a break from competitive games for a few months, I am using this fixture as an opportunity to try out new players and view their potential.
"Greece are currently 10th in FIFA’s World Rankings, and will provide a very difficult opposition for us. Players like Samaras, Gekas, and Karagounis are extremely talented. I am looking forward to setting our squad up for this challenge."
The 26-man panel will be reduced to 23 when the squad meet up on November 11.
