Returning loanee Kevin De Bruyne and Andre Schurrle will both start the game while Marco van Ginkel will appear in the second half, with Mourinho promising to play two teams in Bangkok.
Wednesday's fixture gives the Singha All-Stars an opportunity to continue their winning streak after defeating Manchester United over the weekend in the Red Devils' opening pre-season friendly.
"We expect a difficult game from the Singha All-Stars, they are in good condition, but nobody wants to lose the game," Mourinho told reporters.
"We will play two teams, one in the first half and one in the second. Everybody will play 45 minutes and they have to give strong performances."
De Bruyne is the only player to feature in both teams for Wednesday's game and he starts in midfield alongside Frank Lampard and Michael Essien.
