The former Netherlands international underwent surgery on Thursday and will hand over his duties to assistant boss Ernest Faber while he makes his recovery.
A statement released by PSV on Thursday said: "Head coach Phillip Cocu this morning underwent a successful back operation in which a tumor was removed from his back. It looks very likely to be a benign variant.
"The executive medical team and the medical staff of PSV are satisfied with the conduct of the operation but will not available for further comment."
On Friday, PSV sporting director Marcel Brands will hold a press conference along with a player ahead of Saturday's clash with Groningen.
Cocu has guided the club into second place ahead of that game, six points adrift of leaders Ajax, who have a game in hand.
