The Croatian midfielder was ill and did not feature for Spurs in the 3-1 victory over West Brom at the Hawthorns on Saturday.
But Redknapp has selected Modric, as well as Aaron Lennon, William Gallas and Jermain Defoe, as Tottenham go in search of a vital three points following Rubin Kazan's 4-1 win over Shamrock Rovers.
Spurs must beat PAOK, who are a point ahead of the north London side ahead of the game at White Hart Lane, to move into the top two of Group A.
Elsewhere, Heurelho Gomes starts in goal, while Steven Pienaar continues his recovery from injury.
Young striker Harry Kane partners Defoe in attack as a knee injury rules out Russian international Roman Pavlyuchenko.
