The Yorkshire side achieved a well earned 2-2 draw at White Hart Lane in the first match between the two and are only one of two League One sides still in the competition.
Leeds have not recorded a victory against the Londoners in six league and cup fixtures, however, with their last success coming in the 2-1 Premier League win back in 2001.
Both Neil Kilkenny and Patrick Kisnorbo face a race against time to be fit for the home side, with loanees Shane Lowry, Max Gradel and Gary McSheffrey ineligible.
Tottenham will be without the services of Russian striker Roman Pavlyuchenko due to a groin problem whilst Ledley King is again not expected to feature.
Aaron Lennon and Benoit Assou-Ekotto are also injury related absentees, so youngsters Harry Kane and Dean Parrett will take their places in the squad.
The tie has come too early for Spurs' new front-man Eidur Gudjohnsen to be considered, whilst fellow newcomer Younes Kaboul is cup-tied.
A trip to The Reebok to face Bolton Wanderers in the last 16 awaits the winners.
