The trainer is resigned to losing the talented 25-year-old, but believes the defender should shun Premier League advances in favour of a move to the Blaugrana.
Vertonghen has long been the subject of speculation and a host of teams, including Arsenal and Tottenham, are reportedly vying for his services.
"I hope he will leave for Barcelona. Of course, Tottenham is a great club for Jan, but I think he will fit perfectly at Barca," De Boer told reporters.
The defender revealed that he is set to leave the Amsterdam ArenA after admitting that he played his last home game for the club in the 2-0 victory over VVV.
When asked if this was his last home appearance for the club, the defender was categorical:
"That's almost sure, yes," said the 25-year-old to reporters.
Vertonghen is a product of Ajax's youth academy, and has made over 150 appearances for Ajax, scoring 22 goals.
